Multiplexed affinity purification and thermal dissociation prior to biochip hybridization simplifies uncharacterized sample admixtures, thereby minimizing or eliminating sample interferents, improving hybridization specificity on a microarray detector, and minimizing or eliminating the need for post-hybridization thermal dissociation analysis. An integrated thermo-affinity sample preparation sub-circuit for sample purification and enrichment is described that is consistent with a field-portable form factor and analytical processes. Thermo-affinity sample preparation on model admixtures of varying complexity was efficacious.

Nucleic acid sample purification and enrichment with a thermo-affinity microfluidic sub-circuit

New gel-forming reagents including monomers and cross-linkers, which can be applied to gel-drop microarray manufacturing by using co-polymerization approaches are disclosed. Compositions for the preparation of co-polymerization mixtures with new gel-forming monomers and cross-linker reagents are described herein. New co-polymerization compositions and cross-linkers with variable length linker groups between unsaturated C&#x2550;C bonds that participate in the formation of gel networks are disclosed.

Gel-forming reagents and uses thereof for preparing microarrays

New modifiers were synthesized for incorporation of a methacrylic function in 3'-, 5&#x2032;- and internal positions of oligonucleotides during solid phase synthesis. A modifier was used for synthesis of 5&#x2032;-methacrylated oligonucleotides for preparation of microarrays by a co-polymerization method.

Synthesis of 3'-, or 5&#x2032;-, or internal methacrylamido-modified oligonucleotides

This invention describes a new method of constructing double-stranded DNA (dsDNA) microarrays based on the use of pre-synthesized or natural DNA duplexes without a stem-loop structure. The complementary oligonucleotide chains are bonded together by a novel connector that includes a linker for immobilization on a matrix. A non-enzymatic method for synthesizing double-stranded nucleic acids with this novel connector enables the construction of inexpensive and robust dsDNA/dsRNA microarrays. DNA-DNA and DNA-protein interactions are investigated using the microarrays.
