Provided herein is technology relating to vaccines and particularly, but not exclusively, to compositions, methods, and uses of a mixture of immunogenic vaccine molecules comprising components for targeting the dimeric vaccine molecules to antigen-presenting cells and components for eliciting an immunogenic response, wherein the components for eliciting an immunogenic response preferably comprise at least three variants of an immunogenic protein, such as variants of immunogenic proteins obtained from three or more different strains of a pathogenic organism.

Vaccine molecules

The present invention relates to an immunoglobulin derived single-chain fragment variable (scFv) that broadly binds HLA II molecules and uses thereof. In particular, targeting of an antigen to antigen presenting cells with the HLAII-specific targeting unit provided herein find use in enhancing immune responses after vaccination.

HLA binding vaccine moieties and uses thereof

Provided herein is technology relating to vaccines and particularly, but not exclusively, to compositions, methods, and uses of heterodimer vaccine molecules formed from monomers comprising a targeting unit and a variant antigenic unit joined by heterodimerization units.

Fusion proteins for the treatment of allergic diseases

Recombinant antibody-based molecules that trigger both T-cell and B-cell immune responses are disclosed. The recombinant molecules are comprised by at least one targeting unit and at least one antigenic unit connected through a dimerization motif. Also disclosed are nucleic acid molecules encoding the recombinant antibody-based molecule and methods of treating multiple myeloma or lymphoma in a patient using the recombinant antibody-based molecules or the nucleic acid molecules.

Modified antibody

The present invention relates to disulphide bond stabilized recombinant MHC class II molecules. In particular, the present invention provides a recombinant MHC class II molecule, which comprises: (i) all or part of the extracellular portion of an MHC class II &#x3b1; chain; (ii) all or part of the extracellular portion of an MHC class II &#x3b2; chain; wherein (i) and (ii) provide a functional peptide binding domain and wherein (i) and (ii) are linked by a disulphide bond between cysteine residues located in the &#x3b1;2 domain of said &#x3b1; chain and the &#x3b2;2 domain of said &#x3b2; chain, wherein said cysteine residues are not present in native MHC class II &#x3b1;2 and &#x3b2;2 domains. Methods of producing these molecules in prokaryotic systems and various uses of these molecules form further aspects.

Disulphide bond-stabilized functional soluble MHC class II heterodimers

The present invention relates to a fusion protein comprising a first peptide and a second peptide linked together with a linker, wherein the first peptide is an allergen and the second peptide is a targeting unit and the second targeting unit peptide is a FGL-2 C-terminal peptide according to SEQ ID no 1. Provided herein are also uses of said fusion protein as a vaccine for treating shrimp allergy, as well as a vaccine composition and methods of its production.

A modified immunoglobulin (Ig) molecule incorporates, preferably in one or more non-CDR loops, one or more foreign antigenic peptides such as a ras peptide. The antigen binding site of the immunoglobulin preferably recognises dendritic antigen presenting cells (APCs). The modified Ig can thus be taken up by dendritic APCs and the foreign antigenic peptide presented on MHC II to naive T-helper cells which stimulate cytotoxic T-cells via the production inter alia of IL-2. Modified Igs of the invention can be used to stimulate the immune system which has apparently become tolerant of a mutant protein, e.g., in the case of certain types of cancer, or it could be used for vaccination against viral infections. The modified Ig can be expressed from recombinant host cells from which it is secreted, notwithstanding the presence of the foreign pepide in a loop of the molecule.
