The present invention provides a performance assay that measures the total antioxidant activity of a composition using oxygen uptake in contrast to prior art methods that measure antioxidant capacity by indirectly measuring degradation of a fluorescent compound by following the disappearance of fluorescence. Using the performance antioxidant assay of the present invention, an antioxidant composition having synergistic activity is provided by the present inventors that includes flavonoids such as the flavonol quercetin, mixed tocopherols or tocotrienols, grape skin extract, green tea extract and bush plum. The antioxidant activity of the present composition exceeds 6,000 micromoles Trolox equivalent units per gram using the present invention.

The process of sizing can be accomplished by centrifugation, filtration, ultrafiltration, chromatography, dialysis, selective precipitation, pH adjustment, irradiation, homogenizing, or a combination of such processes. Compounding or mixing two or more different bioactive Factors from aloe at various concentration ratios, each Factor with its own specific bioactivity or bioactivities, to obtain a new bioactive Factor having desired optimal additive or synergistic effects.

Acemannan has direct stimulatory effects on the immune system. An assay for the qualitative and quantitative determination of a bioactive polysaccharide, such as acemannan, in a product, such as an aloe vera-containing product. The method uses a complexing agent to react with the bioactive polysaccharide present, if any, in the product to give a color change. The detected color change is compared with standard color changes generated by reacting the complexing agent with different known amounts of the bioactive polysaccharide, wherein differences between the color changes compared are dependent upon the presence or amount of the bioactive polysaccharide in the product. The assay can be confirmed by detecting a second color change of a different aliquot of the product with a different complexing agent.
