Aggregation-induced emission luminogens useful for imaging β-amyloid peptide and aggregates thereof, pharmaceutical compositions comprising the same, and methods of use and preparation thereof.

Probes for imaging b-amyloid and inhibition of β-amyloid aggregation

A compound comprises a donor and an acceptor, wherein at least one donor ('D') and at least one acceptor (“A”) may be arranged in an order of D-A; D-A-D; A-D-A; D-D-A-D-D; A-A-D-A-A; D-A-D-A-D; and A-D-A-D-A. The compound may be selected from the group consisting of: MTPE-TP, MTPE-TT, TPE-TP A-TT, PTZ-BT-TP A, NPB-TQ, TPE-TQ-A, MTPE-BTSe, DCDPP-2TP A, DCDPP-2TPA4M, DCDP-2TPA, DCDP-2TPA4M, TTS, ROpen-DTE-TPECM, and RClosed-DTE-TPECM. The compound may be used as a probe and may be functionalized with special targeted groups to image biological species. As non-limiting examples, the compound may be used in cellular cytoplasms or tissue imaging, blood vessel imaging, in vivo fluorescence imaging, brain vascular imaging, sentinel lymph node mapping, and tumor imaging, and the compound may be used as a photoacoustic agent.

Luminogens for biological applications

Fluorescent probes for silver ion detection include organic, water-soluble compounds having aggregation-induced emission (AIE) characteristics. The probes can sense or detect silver ions through aggregation or a precipitation reaction between the silver ions and the organic compounds which induces fluorescence. The compounds are acidic, soluble in aqueous phase, and provide low background fluorescence in aqueous solutions.

Fluorescent probes for silver ION detection

Small molecule compounds having aggregation-induced emission (AIE) characteristics include donor-acceptor (D-A) structural NIR fluorophores based on 1,3-bis(dicyanomethylene)indan (BDCI), a strong and steric electron-deficient acceptor. A twisted quasi-double bond (TQDB) links each electron donor with the acceptor. The compounds can be used as NIR-II fluorescent dyes for in vivo imaging. The compounds can conjugate with bioactive molecules, such as peptides, sugars, aptamers and antibodies, to provide specific and active NIR-II fluorescent probes. The compounds can serve as active NIR-II fluorescent probes in many applications, such as, cancer cell-targeted imaging, accurate diagnosis of disease, and image-guided phototherapy.

Multifunctional photoresponsive materials exhibiting aggregation-induced emission and solid-state actuation

A fluorescent probe can include a compound exhibiting aggregation-induced emission (AIE). The probe can be used for selectively staining mitochondria and lipid droplets and detecting esterase activity. The probe includes an acetoxyl group which can be recognized by esterase in an esterase activity assay. As enzyme activity can reflect cell viability, the probe can be useful in cell viability detection. In addition, the probe includes a cationic moiety to target mitochondria and a lipophilic moiety that can target lipid droplets. A calculated log P value of the lipophilic moiety can be larger than 5.

Single AIEgen for multiple tasks: imaging of dual organelles and evaluation of cell viability

A fluorescent compound for acid detection is strongly emissive in the solid state and can aggregate in the presence of an acid. As such, a fluorescent intensity emitted from the compound can be enhanced from an original intensity level in the presence of an acid. The fluorescent compound can emit an enhanced fluorescent intensity in the presence of an ultra-low acid concentration, for example, a concentration of at least about 10M.

AIEgens for cancer cell imaging

A theranostic agent can be used in both photoacoustic imaging (PAI) and photothermal therapy (PTT) applications. The theranostic agent can include a small molecule, organic compound with absorption in the near-infrared (NIR) interrogation window (700-900 nm). The compound can be a biocompatible organic nanoparticle (ONP). The theranostic agent can be effectively used in PAI and PAI-guided PTT applications. The theranostic agent can be administered to a patient to locate a tumor site in the patient using in vivo imaging techniques. Once the tumor site has been determined, the tumor site can be irradiated with near-infrared light to stop or inhibit the growth of the tumor. An exemplary theranostic agent is provided below
