The invention relates to a filamentous bacteriophage, carrying bacterial Lipopolysaccharide (LPS) endotoxin on its surface, for use in treating cancer and in inhibiting angiogenesis, and in a different context, for use in promoting angiogenesis in diseases or conditions in which there is insufficient angiogenesis.

Filamentous bacteriophage as an angiogenesis modulator

The present invention is directed to a filamentous bacteriophage for use in treating a neurodegenerative tauopathy or susceptibility to a neurodegenerative tauopathy and a method of using the bacteriophage for reducing the formation of fibrils of tau protein or for disaggregating pre-formed fibrils of tau protein, such as in a patient suffering from neurodegenerative tauopathy. The filamentous bacteriophage used in the present invention does not display (i) a mammalian cell internalization signal (ii) a &#x3b2;-amyloid antigen or an antibody to &#x3b2;-amyloid, or (iii) a tau protein antigen or an antibody to tau protein.

The present invention is directed to an immunizing composition containing an antigenic product such as a multiple antigen peptide system (MAPS) or a filamentous bacteriophage displaying an A&#x3b2;PP epitope spanning the &#x3b2;-secretase cleavage site of A&#x3b2;PP and a method for inducing an immune response against the &#x3b2;-secretase cleavage site of A&#x3b2;PP using this immunizing composition. Anti-A&#x3b2;P scFvs and single domain antibodies were generated, and when these antibodies were displayed on filamentous phage or as soluble protein molecules, stabilized by the maltose binding protein, they could prevent the fibrilization of A&#x3b2;p 1-40 and disaggregate A&#x3b2;p 1-40 fibrils generated in vitro. The anti-A&#x3b2;p scFv antibodies also stained amyloid neuritic plaques on slices from transgenic mice. More than 90% of mutations found in the p53 protein produce a conformational change in p53 which results in the exposure of an epitope, which is otherwise hidden in the hydrophobic core of the molecule. A single chain antibody (scFv) which specifically recognizes this common mutant epitope in mutant p53 but not in wild type p53 is disclosed. Also described are a DNA molecule encoding the scFv, pharmaceutical compositions comprising the antibody and methods of treatment using the pharmaceutical compositions.
