A method for classifying and counting leukocytes, which comprises steps of: staining cells in a sample obtained from a hematological sample by treatment with a hemolytic agent, with a fluorescent dye; introducing the sample containing the stained cells into a flow cytometer to measure first scattered light, second scattered light different from the first scattered light and fluorescence of the respective cells; obtaining scattered light peak intensities and scattered light widths of the respective cells based on the measured first scattered light, obtaining scattered light intensities of the respective cells based on the measured second scattered light, and obtaining fluorescence intensities of the respective cells based on the measured fluorescence light; classifying the cells into a first group and a second group based on the scattered light peak intensities and the scattered light widths, the first group including leukocytes and second group including platelet clumps; classifying the leukocytes included in the first group into at least lymphocytes, monocytes and granulocytes based on the scattered light intensities and the fluorescence intensities; and counting the classified lymphocytes, the classified monocytes and the classified granulocytes.

An immunoassay comprises the steps of: (a) mixing a whole blood sample with sensitized insoluble carrier particles smaller than erythrocytes to cause an immune agglutination reaction; (b) introducing the resulting immune agglutination reaction mixture including agglutinated particles and unagglutinated particles to a flow cell, irradiating the particles passing through the flow cell with laser light, and detecting scattered lights generated thereby; (c) setting a threshold value for distinguishing unagglutinated particles from agglutinated particles and a threshold value for distinguishing the agglutinated particles from blood cells with regard to intensity of the scattered light; and (d) distinguishing and counting the unagglutinated particles, the agglutinated particles and the blood cells from the scattered lights detected in the step (b), in reference to the threshold values set in the step (c).

A whole blood immunoassay includes the steps of mixing a whole blood sample with sensitized insoluble carrier particles to cause an immune agglutination; diluting the resulting agglutination mixture with an aqueous solution containing an erythrocyte lysing agent to lyse erythrocytes, thereby preparing an assay sample; and determining a degree of agglutination of the assay sample.
