A method of treating a substrate in a pressure vessel that includes the steps: preparing an ion-exchange bath with a bath composition that comprises a polar solvent and a plurality of ion-exchanging ions in a vessel; submersing a substrate having an outer region containing a plurality of exchangeable ions in the bath; pressurizing the bath in the vessel to a predetermined pressure substantially above ambient pressure; heating the bath in the vessel to a predetermined temperature; and treating the substrate for a predetermined ion-exchange duration at the predetermined pressure and temperature such that a portion of the plurality of exchangeable ions is exchanged with a portion of the ion-exchanging ions. The substrate can consist essentially of a glass, glass-ceramic or ceramic substrate composition, and the predetermined ion-exchange duration, temperature and pressure can each be selected based at least in part on the substrate composition and the bath composition.

Antimicrobial and strengthened-glass articles through pressurized ion exchange

In the present specification and claims, three modified cell culture substrates are disclosed for protecting peptide mimetic surfaces used in cell culture technology. The methods are able to retain the functionality of the bioactive species conjugated to the surface. In particular, a vitronectin peptide fragment Ac-Lys-Gly-Gly-Pro-Gln-Val-Thr-Arg-Gly-Asp-Val-Phe-Thr-Met-Pro-NH2 was able to facilitate growth and proliferation of undifferentiated human embryonic stem cells after stabilization using 3 different modified cell culture substrates for of protection against gamma irradiation. The modified substrates disclosed in this invention are (i) covalent attachment of the antioxidant molecules (via blocking step); (ii) The use of coatings consisting of anti-oxidants and stabilizers comprising glycoprotiens, sugars, carbohydrates, poly(amino acids), peptides and hydrophilic polymers; and (iii) a third method wherein the same methionine contained in the bioactive sequence used to facilitate growth and proliferation of human embryonic stem cells was also used as a sacrificial coating to protect the same conjugated sequence against damage by gamma irradiation.

Disclosed herein are functionalized cationic peptide monomers: peptide chains of two or more positively charged amino acids chosen from lysine or arginine, or derivatives thereof, which are functionalized, meaning that they are bound to one or more polymerization moieties. The functionalized cationic peptide monomers can be described by the formula Z-Xaa-Zwherein Z and Zare polymerization moieties and n1 is an integer of 0 or 1; Xaa is each independently an amino acid Lys or Arg and n is an integer from 2 to 10 and wherein at least one Xaa amino acid of Xaa is Lys, wherein the carboxyl terminus of the amino acid sequence is amidated. Functionalized cationic peptide monomers can be combined and polymerized to form cell culture surfaces.

Synthetic surfaces capable of supporting culture of undifferentiated human embryonic stem cells in a chemically defined medium include a swellable (meth)acrylate layer and a peptide conjugated to the swellable (meth)acrylate layer. The swellable (meth)acrylate layer may be formed by polymerizing monomers in a composition that includes hydroxyethyl methacrylate, 2-carboxyehylacrylate, and tetra(ethylene glycol)dimethacrylate. The conjugated peptide may include an amino acid sequence of XaaProGlnValThrArgGlyAspValPheThrMetPro, where n is an integer from 0 to 3 and where Xaa is any amino acid. Synthetic surfaces capable of supporting culture of undifferentiated human embryonic stem cells in a chemically defined medium include a swellable (meth)acrylate layer and a peptide conjugated to the swellable (meth)acrylate layer. A method for producing a cell culture article having a synthetic polymer layer for incubating with cells includes diluting one or more (meth)acrylate monomers in a solvent and dispersing the diluted monomers on a surface of the cell culture article. Some or substantially all of the solvent is removed and the monomers are then polymerized on the surface of the article to form the synthetic polymer layer attached to the surface of the article.
