A synthetic RNA catalyst capable of cleaving an RNA substrate, the catalyst comprising a substrate binding portion and a &ldquo;hairpin&rdquo; portion. The invention also provides an engineered DNA molecule and a vector, each comprising a DNA sequence coding for an RNA catalyst according to the invention. The invention further comprises host cells transformed with the vectors of the invention which are capable of expressing the RNA catalyst. Finally, the invention provides a method of cleaving an RNA substrate which comprises contacting the substrate with a synthetic RNA catalyst according to the invention.

Synthetic catalytic RNAs, i.e. ribozyme, including a hairpin portion, binding sites for binding to a human papilloma virus after viral base 419 and 434, respectively, and cleavage sites for cleaving the virus at the binding sites have been constructed.

Human papilloma virus inhibition by a hairpin ribozyme

A method for the identification by modeling of synthetic catalytic RNAs and ribozymes thereof are disclosed including a hairpin portion and binding sites for cleaving an RNA substrate at a site which has a GUA sequence following the site of cleavage. Synthetic catalytic RNA has been modeled from negative strands of satellite RNA from the group consisting of chicory yellow mottle virus (sCYMVI) and arabis mosaic virus (sArMV).
