A device for analyzing biological samples comprises first, second, third, and fourth layers. The first layer comprises a sample chamber in which a sample is positioned. The second layer comprises first, second, and third channels. A third, porous layer is positioned between the first layer and the second layer. A fourth layer composed of a substantially liquid-impermeable material is positioned between the second layer and the third layer. The fourth layer includes first and second pass-through channels that are aligned with the first and second channel, respectively. Fluids that flow in the first and second channels pass through the pass-through channels and diffuse into the sample chamber, establishing a chemical concentration gradient therein. A gas in the sample chamber can diffuse through the third and fourth layers and interact with a fluid flowing in the third channel, establishing a gas concentration gradient in the sample chamber.

Disclosed herein are methods for tracking solutions, (e.g., reaction conditions in solutions). In some embodiments, the method comprises: contacting a first lanthanide-chelator complex to a first solution to generate a first barcoded solution, wherein the first lanthanide-chelator complex comprises a first lanthanide chelated by a first chelator; contacting a second lanthanide-chelator complex to a second solution to generate a second barcoded solution, wherein the second lanthanide-chelator complex comprises a second lanthanide chelated by a second chelator; mixing the first barcoded solution and the second barcoded solution to form one or more mixtures; and identifying the first lanthanide ions in the mass spectrum and the second lanthanide ions in the mass spectrum to track the condition of each of the one or more mixtures.

Digital microfluidic systems and methods for droplet manipulation

The present invention relates to methods of detecting one or more targets of interest in a sample. In one instance, the target can be correlated to an active infection (e.g., by a virus and/or a bacterium). Methods can include treating the sample with a dissociation agent, thereby releasing the target of interest for more accurate detection (e.g., by use of a sedimentation-based centrifugal microfluidic devices). Also described herein are microfluidic devices and systems for use with a dissociation agent.

In one example, a method includes generating complexes of silica beads and nucleic acids in a lysis buffer, transporting the complexes through an immiscible fluid to remove interfering compounds from the complexes, further transporting the complexes into a density medium containing components required for PCR where the nucleic acids disassociate from the silica beads, and thermocycling the contents of the density medium to achieve PCR. Embodiments of the present invention include methods for conducting agglutination assays using sedimentation. Aggregates may be exposed to sedimentation forces and travel through a density medium to a detection area. A centrifugal force may then be applied to transport the beads through a density media to a detection region for measurement by a detection unit.
