The present invention relates to monolithic intravaginal rings comprising progesterone, methods of making, and uses thereof. The intravaginal rings comprise progesterone, a polysiloxane elastomer, and a pharmaceutically acceptable hydrocarbon or glycerol esters of a fatty acid.

The present invention is directed to methods of reducing symptoms associated with an abrupt reduction in an endogenous or exogenous chemical in subjects, the methods comprising: (a) administering to the subject a single dosage form comprising an active agent, wherein the release rate of the active agent from the single dosage form tapers throughout the administration; and (b) removing the single dosage form from the subjects after the release rate of the active agent is at or below a terminal symptom threshold level.

Provided herein is a novel and useful device and method for locally delivering and controllably releasing oxybutynin in the cervical region of a female. A device of the invention comprises a ring comprising trifluoropropylmethyl/dimethyl siloxane elastomer. A pharmaceutical composition comprising oxybutynin and an excipient is placed within a bore located in the ring, wherein the bore runs from the surface of the ring into the ring. The ring has a sufficient size such that it can be inserted into the vaginal canal of a female. A cap comprising is placed over the bore at the surface of the ring in order to contain the pharmaceutical composition within the bore. When the ring is inserted into the vaginal canal, the trifluoropropylmethyl/dimethyl siloxane elastomer controllably releases and locally delivers a therapeutically effective amount of oxybutynin to the detrusor muscle to treat the female's urinary incontinence.

A novel and useful apparatus for delivering an anesthetic locally to the cervical region of a female is provided, wherein the apparatus comprises a ring having a surface and at least one depression on the surface. An anesthetic composition comprising an anesthetic agent and an excipient are placed within the at least one depression. The ring is comprised of a pharmaceutically acceptable inert material that is biocompatible. Furthermore, the ring is of sufficient size such that it can be inserted into the vaginal canal of a female. In the vaginal canal, the anesthetic agent is immediately released from the anesthetic composition and directly anesthetizes the cervical region.
