Disclosed herein are methods of protein quantification and normalization using haloalkylated tryptophan fluorescence. Complex protein samples, i.e., samples that each contain 1,000 or more distinct proteins, from diverse sources that do not have common protein profiles are treated with a halo-substituted organic compound (i.e. haloalkane) that reacts with tryptophan residues to form fluorescent products. Irradiation of the samples with ultraviolet light and the detection and quantification of the resultant fluorescent emissions from all proteins in each sample are then used to obtain comparative values for total protein content among the various samples. The values thus obtained are found to be valid indications of comparative total protein content, despite the fact that the tryptophan levels vary widely among the various proteins in any single sample and the samples, due to the diversity of their origins, tend to differ among themselves in the identities and relative amounts of the proteins that they contain. Protein samples are also normalized to correct for differences in sample dilution, sample loading, and protein transfer inconsistencies, by using stain-free detection of total protein in each of the samples, or detection of subsamples within each sample.

A gel electrophoresis cassette () is disclosed which includes two plates () and at least one seal () which separates these plates. The seal () is annular; it may be positioned essentially in the region of the outer edge of the plates (). For performing an electrophoresis in a second dimension following an isoelectric focusing in an first dimension, one of the plates () includes a recess () for inserting a strip holder () having a base plate (), which has a carrier surface () on which an IEF strip () is accommodated. The base plate () includes at least one stop () offset to a lower level in relation to the carrier surface () and a sealing surface (). The strip holder () is insertable into this recess () in such a way that the stop () of the base plate () is applied to the outer surface of the plate () and the sealing surface () presses tightly against the inner surface () of the recess (). In addition, systems and methods for performing a 2-D gel electrophoresis are disclosed.

Cassette, system, and 2-D gel electrophoresis method for separating molecules

A strip holder () is disclosed for accommodating a gel strip () for separating molecules using gel electrophoresis. The strip holder () disclosed is distinguished by including a baseplate (), at least one stop (), which is offset to a lower level in relation to the carrier surface (), and at least one sealing surface (), this stop () being implemented to be applied to counter surfaces () of an electrophoresis chamber, through which offset to a lower level of the stop () the installation depth of the strip holder () carrying a gel strip () into this electrophoresis chamber is determined and the sealing surface () ensuring a sealing installation of the strip holder () carrying a gel strip () into this electrophoresis chamber. Such a chamber () for isoelectric focusing of molecules in gel strips () is distinguished by including such a strip holder (), a frame (), and a cover (). Another chamber in the form of a cassette (), for performing an electrophoresis in the second dimension following the isoelectric focusing, includes two plates () and at least one seal () which separates these plates and is distinguished by including such a strip holder (), the strip holder being sealingly inserted into a recess () in one of these plates (). The first dimension of a corresponding 2-D gel electrophoresis method is also disclosed.
