The present invention provides methods for analyzing a target compound from a biological sample. In one aspect, a method for analyzing a target compound in a biological sample can comprise delivering a biological sample through an affinity column, the affinity column having a binding ligand coupled to a stationary structural support, wherein the affinity column has a high density of the binding ligand per the stationary structural support and wherein the binding ligand has been preselected to cause weak affinity separation zonal retardation of the target compound from the biological sample forming a target compound fraction and a biological sample fraction and detecting the target compound by mass spectrometry.

Methods and materials are disclosed relating to an improved method for amplifying a signal in a diagnostic assay for a nucleic acid, comprising the steps of providing an amplification polymer bound to a nucleic acid analyte, wherein the amplification polymer comprises a plurality of amine groups; binding amine groups on the amplification polymer with a detectable label complex; and reacting under high salt conditions an acetylating compound with amine groups not bound with a detectable label complex.

The present invention provides methods for separating proteins from a protein mixture. In one aspect, a method for separating a high concentration protein mixture into a bound protein fraction and a flow-through protein fraction can include delivering a protein mixture through an ion exchange column at a fixed pH and a fixed salt concentration. The fixed pH and the fixed salt concentration have been preselected to cause separation of the protein mixture into a bound protein fraction and a flow-through protein fraction. In this case, the bound protein fraction binds to the ion exchange column and the flow-though protein fraction flows though the ion exchange column. The method can further include receiving the flow-through protein fraction from the ion exchange column separate from the bound protein fraction, wherein either the bound protein fraction or the flow-through fraction contains a protein of interest.

Protein separation via ion-exchange chromatography and associated methods, systems, and devices

The present invention provides methods for separating proteins from a protein mixture. In one aspect, a method for separating a high concentration protein mixture into a bound protein fraction and a flow-through protein fraction can include delivering a protein mixture through an ion exchange column at a fixed pH and a fixed salt concentration. The fixed pH and the fixed salt concentration have been preselected to cause separation of the protein mixture into a bound protein fraction and a flow-through protein fraction. In this case, the bound protein fraction binds to the ion exchange column and the flow-through protein fraction flows though the ion exchange column. The method can further include receiving the flow-through protein fraction from the ion exchange column separate from the bound protein fraction, wherein either the bound protein fraction or the flow-through fraction contains a protein of interest.

High purity protein A is recovered from protein A preparations by passing a protein A preparation over an ion exchange column so that protein A will bind to a cation exchange resin. The loaded column is washed with a wash buffer solution having about the same pH as the protein A preparation and an increased mineral salt concentration to elute certain impurities but not the protein A. The protein A is eluted from the washed column by eluting the column with an elution buffer solution having a pH higher than the wash buffer solution and mineral salt concentration substantially no higher than the wash buffer solution. The protein A can be easily recovered from protein A preparations that do not use any human or animal components. The protein A purified by the invention is suitable for all common uses of protein A, but is especially well-suited for human therapeutic applications.

A method of reducing the chance of disease resulting from mononuclear cells in a body attacking desirable cell antigens in the body, such as antigens expressed by normal body cells or desirable engrafted cells, comprises reducing the number of mononuclear cell in the body that are likely to attack such desirable cell antigens. This is done by obtaining cell derived antigens subject to attack by mononuclear cells in the body and also obtaining mononuclear cells to be placed in the body. The mononuclear cells may be from a donor or may be removed from the subject body. The mononuclear cells are reacted, in vitro, with the cell derived antigens, such as by passing the mononuclear cells over a layer of fibroblast cells from the body expressing the desirable antigens, and recovering the mononuclear cells that do not bind to the antigens. These recovered cells are then infused into the body.

A method of separating charged molecules having different numbers of charges thereon includes the step of passing the molecules to be separated through ion-exchange material such as an ion-exchange chromatograpy column in normal manner so that molecules to be separated bind to the ion-exchange material. Displacer molecules are then passed through the ion-exchange column to selectively release certain of the molecules to be separated from the ion-exchange material, while others of the molecules to be separated remain bound to the material. Rather than the displacer molecules having charges of a similar sign to the molecules to be separated so that the displacer molecules bind to the ion-exchange material in place of certain of the molecules to be separated thereby releasing them, the displacer molecules have charges of the same sign as the ion-exchange material and opposite the sign of the molecules to be separated so that certain of the molecules to be separated bind to the displacer molecules in place of the ion-exchange material and are thus released from the ion-exchange material and flow from the ion-exchange material with the diplacers. Displacers of various numbers of charges may be sequentially passed through the ion-exchange material in ascending number of charge order to thereby selectively release molecules to be separated. The last remaining molecules to be separated may be released selectively with displacer molecules or, may be released in known manner such as by elution.
