Provided are compositions, systems, and methods useful for effecting gene editing in eukaryotic cells. Compositions include plasmids that encode one or more viral fusion proteins in which one or more viral proteins are fused with an aptamer-binding protein. Compositions also include plasmids that encode a non-viral nucleic acid sequence, wherein the non-viral nucleic acid sequence encodes a CRISPR system component. In some instances, the non-viral nucleic acid sequence also includes an aptamer sequence. The plasmids can be used to generate viral particles, including lentivirus-like particles that contain a viral fusion protein and a non-viral RNA sequence. Systems of producing such viral particles are provided. Also provided are methods of using the viral particles of the disclosure to effect gene editing in eukaryotic cells.

Lentiviral-based vectors and related systems and methods for eukaryotic gene editing

Provided herein are artificial lung organoids. The artificial lung organoids may include an epithelial cell layer comprising mammalian lung epithelial cells, a stromal cell layer comprising mammalian lung fibroblast cells and an endothelial cell layer comprising mammalian endothelial cells. The artificial lung organoids may optionally include a porous membrane between said epithelial cell layer and said stromal cell layer and/or between said stromal cell layer and said endothelial lung cell layer.

Multi-layer airway organoids and methods of making and using the same

Provided herein is a delivery system, including: (a) an optical sensor configured to detect data useful to create a map of a bodily surface; and (b) a printer operatively associated with the optical sensor and configured to deliver compositions (optionally including cells) to the bodily surface based upon the data or map. A method of making an organ or tissue comprises: (a) providing a first dispenser containing a structural support polymer and a second dispenser containing a live cell-containing composition; (b) depositing a layer on said support from said first and second dispenser, said layer comprising a structural support polymer and said cell-containing composition; and then (c) iteratively repeating said depositing step a plurality of times to form a plurality of layers one on another, with separate and discrete regions in each of said layers comprising one or the other of said support polymer or said cell-containing composition, to thereby produce provide a composite three dimensional structure containing both structural support regions and cell-containing regions. Apparatus for carrying out the method and composite products produced by the method are also described.

Integrated organ and tissue printing methods, system and apparatus

The invention is directed to methods of inducing cell recruitment and tissue regeneration at a target site in a subject. It is also based, in part, on the discovery that a subject's own biologic resources and environmental conditions can be used for in situ tissue regeneration and thereby reduce or eliminate the need for donor cell procurement and ex vivo manipulation of such donor cells. Methods are disclosed for recruitment of a subject's own stem cells to a target region by inducing a sustained positive pressure at a target site, such as the kidney, thereby increasing the number of pluripotent cells capable of differentiating to regenerate the target tissue.

Provided herein is a method of forming a ventricular support device for a diseased heart, including providing imaging data of the diseased heart, forming a three-dimensional (3D) heart model based on the imaging data, providing strain data including a plurality of strain estimates for at least one segment of the diseased heart, mapping the plurality of strain estimates onto corresponding portions of the 3D heart model to form a 3D diseased heart model, based on the 3D diseased heart model, forming a model of the ventricular support device configured to surround at least a portion of the diseased heart and provide support based upon said strain estimates, and converting the model of the ventricular support device to a digital file useful for directing a 3D printer device to print said ventricular support device for said diseased heart. Also provided is a customized ventricular support device so produced.

The present application relates to biomimetic three-dimensional (3D) scaffolds, constructs and methods of making the same. The three-dimensional scaffold can include a sacrificial internal cast and a durable external scaffold material, wherein the durable external scaffold material comprises a biocompatible material which completely surrounds the sacrificial internal cast and wherein the sacrificial internal cast be removed to yield a branching 3D network of hollow, vessel-like tubes that substantially mimics a native tissue or organ.

Vascular cast-based scaffolds and methods of making the same

Provided herein is an in vitro model of the blood brain barrier. In some embodiments, the model includes: an endothelial cell layer, and brain tissue layer comprising neuronal cells, and optionally one or more of astrocytes, pericytes, oligodendrocytes, and microglia. In some embodiments, the model further comprises a porous membrane between said endothelial cell layer and the neuronal cell layer. The stem cells may be differentiated into an osteogenic, chondrogenic, adipogenic, endothelial, neurogenic or myogenic lineage.
