The invention discloses a method for sanitization of a bioprocess chromatography column, comprising the steps of: a) providing a bioprocess chromatography column with a packed bed of separation resin particles and a movable adapter, b) with the adapter in contact with the packed bed, conveying a sanitization fluid through the packed bed to a column outlet; c) raising the adapter to provide a gap between the packed bed and the adapter, d) lowering the adapter to close the gap; and e) conveying an equilibration liquid through the packed bed to a column outlet.

Method for sanitization of a chromatography column

The present invention relates to a process for the purification of antibodies from one or more impurities in a liquid, which process comprises contacting said liquid with a first chromatography resin comprised of a support to which multi-modal ligands have been immobilised to adsorb the antibodies to the resin, wherein each multi-modal ligand comprises at least one cation-exchanging group and at least one aromatic or heteroaromatic ring system; adding an eluent to release the antibodies from the resin; and contacting the eluate so obtained with a second chromatography resin. In one embodiment, the ring-forming atoms of the aromatic or heteroaromatic entity are selected from the group consisting of C, S and O, and the cation exchanging group is a weak cation exchanger.

Process for the purification of antibodies

The present invention relates to an IgG-binding compound, which more specifically has affinity for human IgGs of &#x3ba;-type and functional derivatives thereof. More specifically, the compound according to the invention comprises an N,N-alkylated urea moiety located between an aromatic part and another part, which is a linear or cyclic substituted or unsubstituted aliphatic group. The compound binds to a pocket-shaped binding site present on all human IgG &#x3ba;-Fabs, which site is located between the two domains (CH1 and CL) of its constant part. Accordingly, the compound according to the invention is a ligand for human IgGs of &#x3ba;-type, and consequently, the invention also relates to a separation matrix for affinity chromatography, which matrix comprises said compound, as well as to other uses of the compound.

Use of urea variants as affinity ligands

The present invention relates to a chromatographic adsorbent for selectively adsorbing IgG, comprising the following formula and its corresponding enol-form, wherein X represents O, S, or NH; Rrepresents H, Calkyl, Calkoxy, Calkoxy-Calkyl, Ar, &#x2014;C(O)NHR, &#x2014;C(O)&#x2014;Ror halo; Rrepresents H, Calkyl or halo; Rrepresents H, Calkyl, Calkoxy, Calkoxy-Calkyl or Ar; n represents 0, 1, 2 or 3; Y represents a carrier. The present invention also relates to a method of producing said adsorbent as well as use thereof for separating substances by affinity chromatography.
