The present invention relates to a microfluidic assay system and associated reading device, as well as the individual components themselves. The present invention also relates to methods of conducting assays, using a disposable system and associated reading device, as well as kits for conducting assays.

Assay devices and methods

Disclosed is an assay result reading apparatus for use with an assay in which a detectable substance tends to accumulate within a detection zone of the assay, the reading apparatus comprising: a housing or baffle, having a window therein; a light source which emits light through the window so as to illuminate the detection zone of the assay; and a light detector to detect the amount of light reflected and/or transmitted by the detection zone, which amount is at least partly dependent on the amount of detectable substance accumulated in the detection zone; wherein the shape of the window is adapted to render the reading apparatus less sensitive, preferably insensitive, to minor mis-positioning of the detection zone relative to one or more of the window, the light source and the light detector.

Reading of assays

A device and a method for analyzing an analyte in a liquid sample are provided. The device comprises a detecting chamber included in a detection device for containing a detecting element; a liquid sample transferring chamber; and a film with small pores disposed between the detecting chamber and liquid sample transferring chamber. When the detection device is inserted into a liquid sample collecting chamber, the liquid sample in the collecting chamber enters the liquid sample transferring chamber but can not enter the detecting chamber via the film with small pores. When a piston is inserted into the liquid sample transferring chamber, the piston forces a part of liquid sample in the liquid sample transferring chamber to enter the detecting chamber via the film with small pores. Using the device and the method can determine an analyte in a sample quantitatively and can complete the detection in one step.

Device and method for analyzing analyte in liquid samples

An assay result reading device for reading the result of an assay performed using a liquid transport carrier may include at least one light source capable of emitting light incident upon at least one of two or more spatially separated zones of the carrier, a photodetector so positioned as to be capable of detecting light emanating from each of the two said zones and generating signals representing the presence or absence of a fluid sample in the respective zone, and a computation circuit. The computation circuit may be responsive to the signals to calculate a flow rate for a fluid flowing along the carrier, compare the calculated flow rate to upper and lower limits, and reject the assay result if the calculated flow rate is outside the upper and lower limits.

Flow sensing for determination of assay results

Disclosed, in one aspect, is an assay result reading device for reading the result of an assay performed using a test strip, the device comprising: a light source or sources, said light source/s emitting light incident upon at least two, spatially separated zones of the test strip; and a photodetector which detects light emanating from each of the two said zones; in a further aspect is disclosed an assay result reading device for reading the result of an assay performed using a test strip, the device comprising: at least one light source incident upon a zone of the test strip; and at least two photodetectors both of which are able to detect some of the light emanating from the zone of the test strip illuminated by the light source.

Optical arrangement for assay reading device

A device for determining the result of an assay may include a computation circuit, responsive to a signal representing the amount of an analyte or the rate of accumulation of an analyte, to: compare the signal to a first threshold; compare the signal to a second threshold, the second threshold being less than the first threshold; generate an output signal if the signal exceeds the first threshold or the signal is less than the second threshold, the output signal indicative of a first result if the signal exceeds the first threshold, or, alternatively, the output signal indicative of a second result if the signal is less than the second threshold; and terminate the assay if the signal exceeds the first threshold or the signal is less than the second threshold.

A method for determining the time of maximum fertility in the mammalian ovulation cycle, for the purpose of assisting conception, wherein testing is conducted over a period of days in the current ovulation cycle on samples of body fluid obtained from an individual human subject to detect an elevated concentration of first analyte, such as luteinising hormone (LH) indicative of the event of ovulation, and additionally testing is conducted over a period of days in the current ovulation cycle on samples of body fluid obtained from the individual subject to detect an elevated concentration of a second analyte, such as estradiol or a metabolite thereof, especially estradiol-3-glucuronide (E3G), to provide advance warning of ovulation.
