The present invention relates to a proNGF mutant and to uses thereof, in particular the use of a proNGF mutant for producing human beta-NGF. The present invention discloses a method of preparing a biologically active human beta-NGF from an inactive insoluble proNGF mutant. A proNGF mutant of the invention is substituted by any amino acid but not Arg or Lys at the native protease cleavage site RSKRat least at positions Rand Kcorresponding to positions 101 and 103 of the human wildtype proNGF sequence.

ProNGF mutants and uses thereof in the production of beta-NGF

The invention relates to an expression system comprising a prokaryotic cell which contains a polynucleotide construct that encodes a T7 RNA polymerase under the control of a T7 promoter and under the control of an inducible promoter. The invention also relates to an expression vector which contains a gene that encodes a protein to be expressed under the control of a T7 promoter, characterized in that said expression vector comprises a plasmid stabilization system.
