The present teachings relate to methods, systems, and kits for the preparation, purification and/or analysis of a glycan or glycoconjugate, and specifically to a magnetic bead based sample preparation protocol. In some aspects, the sample preparation protocol can provide for glycoconjugate capture, glycan release, fluorescent derivatization, and glycan purification for subsequent capillary electrophoresis, liquid chromatography, or other glycoanalytical method using magnetic beads containing negatively charged carboxyl groups extending from the surface of the magnetic beads.

Glycan sample preparation

A separation device receives a known or unknown glycan that is co-injected with three different oligomers maltooligosaccharide (MOL). A detector measures the separated glycan and the separated three different oligomers as intensity peaks that are a function of migration time. The migration times of a plurality of other oligomers of MOL are calculated from the migration times of the three different oligomers. Glucose unit (GU) values for the intensity peaks of the separated glycan are calculated by comparing their migration times to the calculated migration times of the plurality of other oligomers of MOL. The processor identifies the structure of the glycan by comparing the calculated GU values of the intensity peaks of the separated glycan to a database of GU values for known glycan structures.

Flow capture device and method for removing cells from blood The current invention discloses a blood treating and/or purifying device for removing circulating pathogens, preferably pathogenic cells, more preferably circulating tumor cells from the blood of a patient, a method of producing such a device and method to treat cancer and other diseases caused by virus infection, bacterial infection and parasites infection as well as autoimmune disorders. The described method is an extracorporeal medical therapy, thus can be done also in a hemodialysis system. The current invention also describes a device and an in-situ production method of preparing the device to remove CTC and other pathogens i.e. virus, bacteria or parasites from the bloodstream.

The present teachings relate to methods, systems, and kits for the preparation, purification and/or analysis of a glycan or glycoconjugate, and specifically to a magnetic bead based sample preparation protocol that can enable full automation and reduced sample preparation time relative to current methods of glycoanalysis. In some aspects, the sample preparation protocol can provide for glycoconjugate capture, glycan release, fluorescent derivatization, and glycan purification for subsequent capillary electrophoresis, liquid chromatography, or other glycoanalytical method without requiring time-consuming sample preparation steps such as centrifugation or vacuum-centrifugation.

A method or platform for monoclonal antibody based biomarker discovery is disclosed. The method according to the invention provides for the integration of analyte collection, hybridoma screening and nanovolume integrated mass spectrometry (NVIMS) to achieve a robust screening system that is capable, for example, of cutting 4-6 years off of the classical biomarker discovery and development process. The invention provides a platform for the rapid, high-throughput production, isolation and characterization of, e.g., disease specific biomarkers together with highly specific monoclonal antibodies. The method of the invention has a variety of applications such as, but not limited to, drug testing, biohazard applications, ecological applications, physiological applications and/or pathology screening applications. The method of the invention is also capable of being performed or used as or with a high-throughput screening process or system of the invention.

Monoclonal antibody based biomarker discovery and development platform

A cost-effective multi-channel capillary gel-electrophoresis system for highly efficient, high speed, high throughput, carbohydrate analysis. In one aspect of the present invention, a high-performance capillary electrophoresis analyzer system has been optimized for carbohydrate analysis application. The system uses a multiplexed/time-staggered fluorescence type detection mechanism, with an integrated fiber optic array-based technology and a novel disposable gel-cartridge. Twelve carbohydrate samples are automatically injected, electrophoretically separated, detected and analyzed simultaneously by using a multiple usage and disposable multi-capillary gel-cartridge. Using commercially available labeling agent such as APTS (representing 8-Aminopyrene-1,3,6-trisulfonic acid, trisodium salt) as an indicator, the capillary electrophoresis-based carbohydrate system/analyzer provides high resolving power in 2-15 minutes of separations. The system can hold a total of 96 samples, which can be automatically analyzed within 30-60 minutes. This affordable fiber optic based fluorescence detection system can be used in laboratories for high speed carbohydrate sequencing & screening applications.

Reactors and methods for miniaturized reactions having enhanced reaction kinetics. In particular the subject matter is directed to chemical and biological reactions conducted in a nanoporous membrane environment. The subject matter contemplates methods for modifying the kinetics of reactions and devices for conducting reactions having modified kinetics. The subject matter also provides systems for rapid miniaturized reactions. Further the subject matter includes methods and kits for conducting a reaction with enhanced throughput and methods of conducting miniaturized, high throughput analyses of reaction products, and the like. Reactions performed on or within a nanoporous membrane exhibits improved kinetic characteristics.

A proteome analyzer includes a separation cassette having a first dimension separation compartment for separation of protein samples by isoelectric focusing and a second dimension separation compartment for separation of protein samples by SDS-polymer network electrophoresis. The first dimension compartment is a reservoir in which a porous material having capillary channels is disposed. The protein samples are disposed in the capillary channels and, in the presence of a pH gradient, are focused spatially by isolectric focusing upon application of an electric field. The second dimension compartment consists of two glass or plastic plates separated by a separation medium. The separation medium is an ultra-thin layer of a low concentration linear polymer supported by an inert matrix. The spatially focused protein samples are contacted with the separation medium in the presence of an electric field to initiate second dimension separation. The migrating SDS-protein complexes are fluorescently labeled by a dye in the separation medium during second dimension separation, and are detected by an illumination and detection system as they pass through a detection area. The detected SDS-protein complexes are imaged and displayed by computer analysis as two-dimensional maps representing the proteins in the sample.

Methods for profiling oligosaccharides released from glycoproteins and methods for the electrophoretic separation of high mannose, complex and hybrid oligosaccharides are disclosed. The methods include first providing an analytical sample which includes at least one released oligosaccharide and introducing the sample into an electrophoretic channel containing a separation medium having a pH less than 8 and greater than 2.5. Then applying a sufficiently high electric field causes the released oligosaccharides to migrate and separate within the electrophoretic channel in accordance with their molecular size or charge to mass ratio. Detecting the separated oligosaccharides provides glycoprotein conjugate profiling information. Preferred embodiments include using a separation medium which includes lithium acetate buffer having a pH of about 4.75 and a gel of about 0.4% polyethylene oxide.

Compositions useful as a separation medium in capillary electrophoresis procedure and methods for their use in capillary electrophoresis are described. The compositions include dry linear polyacrylamide which is capable of hydrating to form an aqueous polymeric system capable of filling an electrophoresis capillary column. Also disclosed are methods for using dry polyacrylamide in capillary electrophoresis. The methods include providing a dry polyacrylamide composition, reconstituting the dry polymer to provide a solution of reconstituted polymer, and causing the reconstituted polymer to fill a capillary electrophoresis column.

Rehydratable gels for capillary electrophoresis

The filling of an internally coated capillary with a gel in its polymerized state without damaging the gel. The coating prevents bonding of the gel to the inside of the capillary, The gel comprises up to 6% acrylamide and 0-5% crosslinker. The gel can be advantageously and conveniently used in automated electrophoresis systems for automatic replacement of spent gel.
