The specification describes a composition comprising an improved eukaryotic cell culture medium, which can be used for the production of a protein of interest. TaXULne can be added to the serum-free media or chemically-defined media to increase the production of a protein of interest. Methods for recombinantly expressing high levels of protein using the media compositions are included.

Anti-VEGF protein compositions and methods for producing the same

The specification describes an improved serum-free animal cell culture medium, which can used for the production of a protein of interest. Ornithine, or a combination of ornithine and putrescine can be added to serum-free media or chemically defined media to improve viable cell density, to reduce cell doubling time, and to increase the production of a protein of interest.

Serum-free cell culture medium

Improved methods for large scale production of proteins and/or polypeptides in cell culture is provided. In accordance with the present invention, the method provides for culturing cells that have metabolically shifted. The use of such a method or system allows high levels of protein or polypeptide production and reduces accumulation of unwanted metabolic waste such as lactate. Proteins and polypeptides expressed in accordance with the present invention may be advantageously used in the preparation of pharmaceutical, immunogenic, or other commercial biologic compositions, such as antibodies.

Metabolically optimized cell culture

A method for culturing eukaryotic cells for improved cell culture performance is provided. The method generally comprises propagating or maintaining eukaryotic cells in a defined cell culture medium; wherein the defined cell culture medium is supplemented with asparagine in an amount from about 2.6 mM to about 43.2 mM during early fed-batch cell culture and from about 2.6 mM to about 21.6 mM during late fed-batch cell culture; and maintaining said cells in said asparagine supplemented cell culture medium for at least a portion of the early fed-batch cell culture and at least a portion of the late fed-batch cell culture; wherein the performance of the cell culture is improved by the asparagine supplementation, as compared to a similar method with a lower amount of asparagine supplementation in the early and/or late fed-batch cell culture.
