Provided are compounds compound according to Formula (I), or a pharmaceutically acceptable salt, solvate, coordination complex or prodrug thereof: wherein, Rand Rare independently selected from (C-C) alkyl. The compounds have P2X3 receptor or P2X2/3 receptor antagonist activity and are useful for the treatment of diseases and disorders characterized by activation of those receptors.

Human mast cell activation is modulated by ATP binding to P2-purinoceptors on the mast cell surface. ATP binding to the purinoceptors provides a target for therapeutic intervention for the treatment of disorders characterized by undesireable mast cell mediator release, such as asthma and allergy. Inhibitors of ATP binding to mast cell P2-purinoceptors are useful therapeutic agents for treatments of those disorders. Methods of treatment using agents, and in vitro screening assays for selection of the therapeutic agents, are described.

Screening method for modulation of human mast cell activation

Human mast cell activation is modulated by ATP binding to P2-purinoceptors on the mast cell surface. Modulation of human mast cell activation

The present invention provides methods of altering vagal tone in a patient by administering a therapeutically effective amount of a mediator of P.sub.2x -purinoceptors located on vagal afferent nerve terminals to the patient. Diagnostic applications are also provided.
