An oral drug delivery system comprising a coated tablet having one or more surfaces. The coated tablet further comprises a core and a coating surrounding the core. The core comprises an active ingredient composition comprising at least one active ingredient and a pharmaceutically acceptable excipient and a swellable composition located in an immediate vicinity of one or more preselected surfaces. The coating comprises a defect wherein said defect is not a passageway through the preselected portion of the coating and operable to be reliably removed fully from the one or more of the preselected surfaces of the tablet upon contact with an aqueous environment, but not removed from at least one of the surfaces.

The coating is operable to be reliably removed fully from the one or more of the preselected surfaces of the tablet upon contact with an aqueous environment, but not removed from at least one of the surfaces.

The present invention is concerned with a system for spatially and temporally programmable delivery of an active agent. It comprises of a core (I), one or more layers (II, IV, V) coated over the core and a preformed hollow space (III). A pharmaceutical composition of anti-tubercular drugs for oral use comprising Rifampicin and/or Isoniazid wherein the bioavailability of Rifampicin and/or other drugs is enhanced. A novel composition for increasing the bioavailability of Non-steroidal Anti-inflammatory Drugs (NSAIDs), particularly those belonging to the category which exhibits its activity by selectively inhibiting cyclooxygenases-II (COX-II) and/or lipooxygenases, is disclosed. The composition is characterized in having clinically significant increased bioavailability when compared to the known compositions of the drugs. The pharmaceutical compositions comprise NSAIDs and Piperine, as herein disclosed. Enhanced bioavailability of NSAIDs results in the reduction of both dosages and dose-related side effects.
