A microbial detection device and methods of use are provided. The device comprises a water-proof pouch that includes a first wall portion, a second wall portion, and a porous membrane filter disposed in the pouch between the first and second wall portions. The filter membrane divides the pouch into first and second compartments. A dry, cold water-soluble gelling agent is adhered to the pouch in the first compartment and an absorbent pad is disposed in the second compartment. A sealable sample port provides access to deposit a liquid into the first compartment.

Described herein is a multilayer article comprising: a. a polymer substrate comprising an abstractable atom; and b. a hydrogel coating thereon wherein the hydrogel coating has a water content of at least 10 wt % and is covalently bonded to the polymer substrate, and wherein the hydrogel coating is derived from an aqueous composition having a pH less than 9.5, the aqueous composition comprising: (a) a hydrophilic monomer selected from at least one of (meth)acrylate or (meth)acrylamide; (b) at least 0.1 wt % of a water-swellable clay; (c) a first initiator, wherein the first initiator is water-soluble and is a Type I photoinitiator; and (d) a second initiator, wherein the initiator is water-soluble and is a Type II photoinitiator; and (e) an acid.

Described herein is a hydrogel-containing multilayer article and methods of making, said hydrogel-containing multilayer article comprising: (i) a polymeric substrate comprising an abstractable atom; and (ii) a cured aqueous coating composition thereon wherein the coating composition comprises: (a) a hydrophilic monomer comprising a (meth)acrylamide, (meth)acrylate, and combinations thereof: (b) at least 2 wt % of a water-swellable clay; (c) a water-soluble type I photoinitiator; and (d) an acid or salt wherein a water insoluble type II photoinitiator is localized at the interface between the hydrogel coating and the polymeric substrate.

The methods include providing a device, separating the first layer from the second layer, adding an aliquot of a sample containing at least one microorganism onto the first or second water-swellable gelling agent to form an inoculated device, laminating the first layer back to the second layer, and incubating the inoculated device. A method of forming a fibrin hydrogel composition is described. The method comprises forming an aqueous solution comprising fibrinogen, fibrin-forming enzyme, and a fibrin hydrogel forming salt. The fibrin hydrogel forming salt concentration is greater than or equal to the threshold concentration to form a fibrin hydrogel. The method further comprises reducing the salt concentration below the threshold concentration to form a fibrin hydrogel. In some embodiments, the aqueous solution further comprises a plasticizer. A fibrin composition is also described comprising a fibrin hydrogel having a fibrin concentration ranging from 0.1 to 10 wt-%; and a fibrin hydrogel forming salt. The fibrin hydrogel forming salt has a concentration less than a threshold concentration to form the fibrin hydrogel. The fibrin hydrogel or dehydrated fibrin hydrogel can be in various physical forms such a sheet, foam, or plurality of pieces. Also described are methods of forming a fibrin article, wound dressings and a method of treatment of a wound.
