The invention provides droplet actuators and droplet actuator cassettes including reagent storage capabilities, as well as methods of making and using the droplet actuators and cassettes. The invention also provides continuous flow channel elements and techniques for using electrodes to manipulate droplets in flowing streams. The invention also discloses methods of separating compounds on a droplet actuator. Various other aspects of the invention are also disclosed.

Droplet actuator and method

Methods of concentrating beads in a droplet and/or loading beads on a fluidic device are provided, including among other things, a method of concentrating beads in a droplet, the method comprising: (a) providing a droplet actuator comprising: (i) an interior droplet operations volume; and (ii) a reservoir exterior to the interior volume; (iii) a droplet established in a liquid path extending from the reservoir into the interior volume; (b) providing magnetically responsive beads in the portion of the droplet which is in the reservoir; (c) magnetically attracting the magnetically responsive beads through the liquid path into the portion of the droplet which is in the interior volume; and (d) forming a droplet comprising one or more of the magnetically responsive beads in the interior volume.

Method of concentrating beads in a droplet

Methods and devices for conducting chemical or biochemical reactions that require multiple reaction temperatures are described. The methods involve moving one or more reaction droplets or reaction volumes through various reaction zones having different temperatures on a microfluidics apparatus. The devices comprise a microfluidics apparatus comprising appropriate actuators capable of moving reaction droplets or reaction volumes through the various reaction zones.

Method and device for conducting biochemical or chemical reactions at multiple temperatures

The invention provides a droplet actuator. The droplet actuator may include a base substrate and a top substrate separated to form a gap. The base substrate may include electrodes configured for conducting droplet operations in the gap; and the top substrate may include a glass substrate portion coupled to a non-glass portion, where the non-glass portion may include one or more openings establishing a fluid path extending from an exterior of the droplet actuator and into the gap. The invention also provides related methods of manufacturing the droplet actuator, methods of using the droplet actuator, and methods of loading the droplet actuator.

The invention provides a method for dispensing liquid, comprising the steps of: (a) positioning a droplet to be dispensed in a gap of an electrowetting device using an electrowetting array; and (b) dispensing the droplet through a hole in a housing or substrate of the electrowetting device. The invention further provides a method for withdrawing liquid comprising the steps of: (a) positioning a droplet to be withdrawn from a gap of an electrowetting device using an electrowetting array; and (b) withdrawing the droplet through a hole in a housing or substrate of the electrowetting device.

Method of loading a droplet actuator

A series of microactuators for manipulating small quantities of liquids, and methods of using these for manipulating liquids, are disclosed. The microactuators are based on the phenomenon of electrowetting and contain no moving parts. The force acting on the liquid is a potential-dependent gradient of adhesion energy between the liquid and a solid insulating surface.

Droplet actuator and methods

The invention provides a droplet actuator comprising a base substrate and a top substrate separated to form a gap, wherein: (a) the base substrate comprises electrodes configured for conducting droplet operations in the gap; and (b) the top substrate comprises a first portion coupled to second portion, where the second portion comprises one or more openings establishing a fluid path extending from an exterior of the droplet actuator and into the gap. The invention also provides related methods of manufacturing the droplet actuator, methods of using the droplet actuator, and methods of loading the droplet actuator.

On-board reagent storage, handling, and dispensing apparatuses and methods for droplet actuator systems are provided. In one embodiment, a fluidic analyzer is provided and includes a droplet actuator including a substrate including a plurality of electrodes configured to actuate a droplet and a reagent storage component in selective fluid communication with the droplet actuator and including a reagent configured to combine with a solvent at a time of use. In another embodiment, a method of conducting fluidic analysis is provided and includes storing a reagent within a reagent storage component in fluid communication with a droplet actuator including a plurality of electrodes configured to actuate a droplet and combining the reagent with a solvent at a time for use within the droplet actuator.

An imaging device consistent with one of numerous embodiments has an opaque planar sheet with a plurality of pinholes defining a photon sieve in the sheet, wherein, the photon sieve comprises at least first and second regions. The first region exhibits a first focal length, a first field of view, a first transmissivity, a first resolution and a first wavelength, and the second region exhibiting a second focal length, a second field of view, a second transmissivity, a second resolution and a second wavelength. At least one of the first focal length, the first wavelength, the first transmissivity, the first resolution and the first field of view is different from the second focal length, the second wavelength, the second transmissivity, the second resolution and the second field of view. This abstract is not to be considered limiting, since other embodiments may deviate from the features described in this abstract.

Method of using actuators for microfluidics without moving parts

An apparatus for inducing movement of an electrolytic droplet includes: a housing having an internal volume filled with a liquid immiscible with an electrolytic droplet; a distribution plate positioned within the chamber having an aperture and dividing the housing into upper and lower chambers; a lower electrode positioned below the lower chamber and the aperture in the distribution plate and being separated from the lower chamber by an overlying hydrophobic insulative layer; an upper electrode located above the upper chamber and the aperture of the distribution plate and being separated from the upper chamber by an underlying hydrophobic insulative layer; and first, second and third voltage generators that are electrically connected to, respectively, the lower and upper electrodes and the distribution plate. The voltage generators are configured to apply electrical potentials to the lower and upper electrodes and the distribution plate, thereby inducing movement of the electrolytic droplet between the hydrophobic layers.
