A method of ligating DNA molecules, wherein the DNA molecules are in a hybrid with an RNA molecule, including the steps of providing DNA molecules that are in a RNA:DNA hybrid with an RNA molecule, and ligating the DNA molecules to each other with a double strand specific ligase.

Strand displacement stop (SDS) ligation

The present invention relates to methods for generating an amplified nucleic acid portion of a template nucleic acid, comprising obtaining said template nucleic acid, annealing at least one oligonucleotide primer to said template nucleic acid, annealing at least one oligonucleotide stopper to said template nucleic acid, elongating the at least one oligonucleotide primer in a template specific manner until the elongating product nucleic acid reaches the position of an annealed oligonucleotide stopper, whereby the elongation reaction is stopped, wherein in said elongation reaction said oligonucleotide stopper is not elongated, and wherein the elongated product nucleic acid is ligated to the 3' end of said oligonucleotide stopper&#x2014;said stopper may also be a primer itself and uses and kits for performing said method.

Nucleic acid transcription method

The present invention relates to methods for generating a labelled nucleic acid from an RNA comprising a 5' protecting group, said method comprises the steps of obtaining a mixture of template strands of nucleic acids, said mixture comprising said RNA and further potentially other nucleic acids without a 5&#x2032; protecting group, annealing at least one oligonucleotide primer to the template strand of said RNA and potentially other nucleic acids, and template sequence dependent extending said primer, thereby obtaining a complementary nucleic acid strand annealed to its template strand, or providing the RNA in duplex with a complementary nucleic acid strand annealed to its template strand, and optionally modifying the extension product of said nucleic acids without 5&#x2032; protecting group either on the 5&#x2032; end of the template strand or on the 3&#x2032; end of the complementary strand, or both, and labelling a complementary nucleic acid of a double stranded nucleic acid not modified, wherein therefore the labelled nucleic acid did have a 5&#x2032; protecting group on the RNA template strand, said 5&#x2032; protecting group being optionally removed after modification, and/or labelling a complementary nucleic acid of a double strand dependent on the presence of the 5&#x2032; protecting group on the complementary nucleic acids template RNA strand by double strand dependent ligation, thereby specifically generating a labelled nucleic acid complementary to an RNA at least originally comprising a 5&#x2032; protecting group.

5' protection dependent amplification

The present invention provides a sensor array device with multiple sensor junctions which have been created through the assembly of two or more differently functionalized surfaces. The functionalizing of the prospective sensor junction areas with sensor compounds occurred when the different surfaces were physically separated from each other before the assembly of the sensor array. By these means, sensor junctions can be built smaller than conventional deposition techniques like printing and photolithography would allow for otherwise. As a consequence, each individual sensor junction contains two potentially different sensor compounds. The sensor array identifies and quantifies different biomolecules.

Biosensor array formed by junctions of functionalized electrodes

We provide a sensor array device for the measurement of mixtures of organic compounds comprising an assembly of sensor half elements which have been functionalized through sensor compounds before the assembly. Each individual sensor of the array contains two sensor compounds which are bound at opposite sensor half elements. The molecular recognition is bi-functional. While the amount of sensor compounds increases linearly, the individual sensors increase with the second power.

The present invention provides a method for amplifying a pool of polynucleotide molecules in a sample, characterized by the steps of a) obtaining a sample or RNA and reverse transcription of entire RNA molecules thus creating full length cDNA or obtaining a sample of full length cDNA, b) tailing the 3' end of the transcribed cDNA with a polynucleotide tail after the 3&#x2032; end, c) amplification of the cDNA using a pair of primers, wherein a first 3&#x2032; primer is specific for the 5&#x2032; end of the cDNA and a second 5&#x2032; primer is specific for the a upstream portion of the polynucleotide tail and the next 1 to 10 nucleotides upstream of the 3&#x2032;polynucleotide tail of the cDNA.

Polynucleotide amplification

A method for the reduction of the complexity of nucleic acid pool(s), comprising: providing a sample with one or more nucleic acid molecules; cutting the nucleic acid molecules by a random and/or sequence independent cutting step thereby obtaining a pool of nucleic acid fragments; and amplifying one or more fragments of said nucleic acid molecules, wherein the one or more fragments constitute at least a fraction of all fragments of the nucleic acid molecules, wherein the amplified or amplifying one or more fragments of said fraction are divided into different subpools, and wherein the fragments of each subpool comprise a common nucleic acid feature.
