The present invention provides methods, systems and kits for performing microwell analysis in sealed small volumes. The microwells of the invention are sealed through the pairing of microwell and bead geometry wherein the diameter of the beads is greater than the diameter of the bottom of the microwells. Loading the beads into the microwells seals samples within an analytical space between the bottom of a nested bead and the bottom of the subject microwell.

Sealed microwell assay

A contact lens assembly includes a proximal layer formed from a gas-permeable material, a distal layer with an optical element, an air cavity disposed between the proximal layer and the distal layer, wherein oxygen concentration across the air cavity is substantially uniform, and an gas transport region surrounding the air cavity for communicating gas between the air cavity and atmosphere. The contact lens assembly may further include a plurality of fins disposed within the air cavity where the plurality of fins has openings therethrough. One or both of the upper and lower surfaces of the air cavity may have an anti-reflection coating formed thereon.

Aerated contact lens assembly

The ability to form and maintain gradients is essential for the study of response of cells to various stimuli. The invention includes devices and methods for the high-throughput, reproducible formation of gradients for the study of living cells. The invention includes microfluidics device with a test chamber having a depth flanked by flow-through channels having a deeper depth. Flow of two different fluids through the flow-through channels results in the creation of a gradient by diffusion across the test chamber having essentially no flow.

Microfluidic device for high-throughput cellular gradient and dose response studies

In a microfluidic system, a magnetic clamp for sealing a flexible microchannel chip includes a base formed from a magnetically-attractable material. The base supports a window for viewing the face of the microfluidic chip. A ring with magnets uniformly distributed around it is disposed over the base. A transparent disk attached to the top of the ring has an inlet and an outlet for introducing and removing a fluid medium into a cavity defined the disk, the window, the center opening of the ring and the base. An elastomer cushion is attached to the inner surface of the disk. The magnetic force between the base and the magnets on the ring compresses the cushion against the microfluidic chip so that the microchannels are sealed against the window with a uniform and reproducible pressure.

Microfluidic system and method for using same

The capillary gun for delivery of ballistic particles to a target includes an inner capillary tube disposed concentrically within an outer capillary tube with the input end of the inner tube connected to a channel through which a continuous flow of high speed helium gas carrying ballistic particles is introduced. The outer capillary tube, which is connected to a vacuum source, has an outlet end that extends slightly beyond the end of the inner tube. A cap placed over the output end of the outer tube has an opening at its center through which the particles exit the device. The vacuum source applies continuous suction to the space between the outer tube and the inner tube, drawing the gas from the output end of the inner tube while the inertia of the accelerated particles causes them to continue in the axial direction through the exit opening for delivery to the target. Multiple particle injectors provide for the concurrent injection of different materials without disruption of the gas flow.
