A separation module and method are disclosed for processing a liquid sample and providing high conversion by operating a single-pass tangential-flow process without a recirculation loop. In one embodiment, the separation module includes three reservoirs and has at least one long, thin channel with a large ratio of channel membrane area to: channel void volume; volume of a sample feed reservoir; and volume of the feed sample. In another embodiment, the separation module includes an external feed reservoir, an external retentate reservoir fluidly and a first vacuum source adapted to apply a first vacuum to the separation element disposed within the housing. The single-pass TFF separation element and single-pass process provides high conversion while operating with relatively low pressure sources. Certain embodiments disclosed herein can be used for the separation of blood.

Method and apparatus for the filtration of biological samples

A functional, porous, interpenetrating polymer network (IPN) includes a first polymer network in the form of a porogenic support fabric (PSF) composed of linear polymers in the form of a pre-formed network comprising a fibrous composite and a second polymer network synthesized, gelated, and/or cross-linked in the presence of the first polymer network to form a system of polymers which have their respective chains held in place by means of permanent physical entanglements produced by the interweaving of the component polymer networks. The IPN is modified by dissolving and dispersing a portion of the PSF fibers, the dispersible fiber network (DFN) to form a pre-designed interconnected pore structure. The resultant porous, supported, second polymer network has convective flow, diffusive flow, and high capacity, and may include functional capture chemistries to provide an adsorptive media for chromatography and filtration of various compounds including biomolecules.

Solid phase synthesis of organic compounds via phosphitylating reagents

A membrane suitable for immobilizing peptides and proteins is disclosed. The membrane is a flexible, polymeric, porous membrane which contains functional groups capable of covalently linking the peptides and proteins. The functional groups can be provided by reacting the membrane itself or a coating thereon with nucleophiles which provide --NH.sub.2, --SH, --OH or --COOH functionality to the membrane surface. Additionally, surfaces containing --NH.sub.2 groups can be further reacted with diisothiocyantes to provide an isothiocyanate functionality having enhanced covalent binding characteristics.

Membranes for solid phase protein sequencing

A process and apparatus for the conversion of anilinothiazolino amino acid derivatives of peptides and proteins for use with conventional sequencers in the overall Edman degradation process. The apparatus comprises a modular unit separate from the sequencer having an independently programmable control and flow actuating and directing means including a source of pressurized nitrogen, a conversion reagent container, a wash solvent container, a reaction vessel, and a plurality of interconnecting tubes and valves controlled so as to regulate the amounts and durations of all fluids entering and leaving said reaction vessel under gas pressure. The process comprises performing the steps of conversion, sample recovery, drying and washing at temperatures below but near to the respective boiling points of both the reagent and the solvent, and under reflux tower conditions. A specific embodiment employs anhydrous methanolic hydrogen chloride for the reagent and dichloroethane methanol (7:3) for the solvent, and both the conversion and the washing are done at 65.degree. C., in a reaction vessel comprising a lower, heated portion, and an upper, cooled portion where the reflux action takes place both during the conversion and washing steps.

A process and apparatus for pulse labelling protein material for high sensitivitiy sequence analysis of protein material by Edman degradation technique comprising an isolated, auxiliary unit for storing a radioactive coupling reagent together with tubes, valves and controls for interrupting the Edman process, for pumping a measure of radioactive coupling reagent into the reaction vessel, a timer for controlling duration of contact between the radioactive reagent, and the protein material to maximize the coupling reaction therebetween, and thereafter reactivating the Edman process to admit nonradioactive coupling reagent to the vessel and to drive the coupling reaction to completion.
