Body-mountable devices are provided to detect the presence or status of a tumor in a body by detecting probes associated with circulating cells of the tumor that travel to subsurface vasculature of the body. The probe enters a tumor and associates with cells of the tumor before the cells metastasize. A wearable body-mountable device can be worn for a protracted period of time to detect the probe associated with circulating tumor cells in the vasculature at low concentrations and/or at low rates. A body-mounted device could detect the presence of such released, tumor-cell-associated probes to determine a presence or status of a tumor in the body.

Method of cell tagging for cancer detection

A method for profiling extracellular vesicles (EV) is provided. The method includes: (a) incubating isolated EV from one or more sources with a phage display library under conditions suitable for forming EV-bound phage, wherein the phage display library comprises one or more types of phage, each type of phage having a displayed proteinaceous display binding moieties capable of recognizing one or more epitopes on the surface of the EV, wherein each type of phage comprises nucleic acids encoding the displayed proteinaceous display binding moieties, and wherein the displayed proteinaceous display binding moieties of each type of phage is different; (b) isolating the EV-bound phage; (c) extracting the nucleic acids from the EV-bound phage; (d) amplifying the extracted nucleic acids; (e) sequencing the amplified nucleic acids to identify specific nucleic acid sequences associated with each type of phage from the library that are enriched from the isolated EV; and (f) comparing the specific nucleic acid sequences (output) with nucleic acid sequences from the phage display library (input) and nucleic acid sequences from a control to identify sequences that can distinguish between EV from the one or more sources.

Methods for profiling extracellular vesicles from combinatorial phage display libraries

Body-mountable devices are provided to detect the presence or status of a tumor in a body by detecting one or more properties of a probe located in subsurface vasculature of the body. A wearable body-mountable device can be worn for a protracted period of time to detect a probe in the vasculature at low concentrations and/or at low rates. A body-mountable device can detect properties of the probe that are indicative of whether the probe has interacted with a tumor of the body and determine the presence or status of a tumor in the body based on such detected properties. Additionally or alternatively, the probe could be introduced into the body as a probe aggregate and released from if the probe aggregate is absorbed by a tumor. The presence of released probes could be detected to determine a presence or status of a tumor in the body.

Enhanced cancer detection through probe modification

A system is provided which includes nanoparticle conjugates configured to bind with a tumor cell, the nanoparticle conjugate comprising a nanoparticle, at least one targeting entity bound to the nanoparticle, and at least one shielding entity that shields at the at least one targeting entity, the nanoparticle, or both; a body-mountable device mounted on an external surface of a living body and configured to detect a tumor cell binding response signal transmitted through the external surface, wherein the tumor cell binding response signal is related to binding of the nanoparticle conjugates with one or more tumor cells; and a processor configured to non-invasively detect the one or more tumor cells based on the tumor cell response signal. Nanoparticle conjugates and methods for use for treating or imaging tumor cells are also provided.
