Preparation of a product comprising stromal vascular fraction cells includes washing human biological material comprising adipose in a container apparatus having an internal filter, which divides an internal containment volume of the container apparatus into a tissue retention volume on one side of the filter and a filtrate volume on an opposite side of the filter, and a mixing device with at least one rotatable mixing member disposed in the tissue retention volume. The washing includes operation of the mixing device to rotate the mixing member through the human biological material within the tissue retention volume, and the washing is followed by digesting washed material within the internal containment volume with added enzyme, centrifuging of the container apparatus to prepare a centrifuged pellet in the filtrate volume, selectively removing material of the pellet and preparing a product with a mixture of stromal vascular fraction cells of removed pellet material and an aqueous suspension liquid.

Method for preparing a product comprising stromal vascular fraction cells

A portable apparatus useful for collection and processing of human biological material, such as adipose or cancellous bone material, to prepare a concentrated product (e.g., stromal vascular fraction). The apparatus has a container with a containment volume with a tissue retention volume and a filtrate volume separated by a filter and with a pellet well for collecting concentrate product in the form of a pellet phase from centrifuge processing. The pellet well is accessible only from above when the apparatus is in an access orientation. Collected pellet phase material may be removed from the pellet well by direct aspiration, without suspending the material in a suspension liquid within the container. The apparatus may include a tissue collector disposed in the disuse retention volume to engage and collect collagen or other stringy tissue. Embodiments of the present disclosure encompass systems and methods for in-situ/in vivo, bottom-up tissue generation for wound repair, repair of tissue defects, and the like. Embodiments of the systems of the present disclosure include modular scaffolds seeded with cells (modular tissue forming units (MTFUs)) for packing a tissue defect, such that these MTFUs are able to fill the wound bed with cells of one or more needed tissue types supported by the modular scaffolding particles.

A portable apparatus for collection and processing of human biological material containing adipose, such as extracted during a lipoplasty procedure, is useful for multi-step processing to prepare a concentrated product (e.g., stromal vascular fraction) or a fat graft composition. The apparatus has a container with an internal containment volume with a tissue retention volume and a filtrate volume separated by a filter and with a tapered portion to a collection volume for collecting concentrate product. The apparatus has fluid suction features including a suction port and a conduit providing fluid communication from the suction port to a location within the filtrate volume within a tapered portion of the internal containment volume, and optionally including a second suction port and a second conduit to a different location in the filtrate volume with the second conduit being configured to permit elevation adjustment within the filtrate volume. Methods include use of the fluid suction features during collection of human biological material and/or post collection.
