A highly controlled and precise system, device and method for tissue and cellular alteration and treatment below or at surfaces with a laser. The present invention is characterized by ultra low levels of collateral damage as defined by physiologically relevant tests that measure tissue viability. The operation of the present invention is based on spectrally confining the interaction between laser energy and a targeted tissue including an essential element for physiologically relevant tests for monitoring tissue viability.

Controlled laser treatment for non-invasive tissue alteration, treatment and diagnostics with minimal collateral damage

A method for deconvolving far-field optical images beyond the diffraction limit includes the use of near-field optical and other scanned probe imaging data to provide powerful and new constraints for the deconvolution of far-field data sets. Near-field data, such as that which can be obtained from atomic force microscopy on a region of the far-field data set in an integrated and inter-digitate way, is used to produce resolutions beyond the diffraction limit of the lens that is being used. In the case of non-linear optical imaging or other microscopies, resolutions beyond that which is achievable with these microscopies can be obtained.

Deconvolving far-field images using scanned probe data

The invention is directed to a scanned probe microscope including one plate allowing for tip scanning and the other allowing for sample scanning, with the optical axis of the scanned probe microscope being free to permit incorporation into standard optical microscopes. The top plate can be hinged onto the bottom plate, or the top plate can simply be placed on the bottom plate and a rough approach is caused by a dc motor or other mechanism which will enable the two flat plate scanners to have a large z range. In another embodiment, the microscope includes three plates which allow sample scanning, tip scanning and two tips to be operational at the same time. A microscope in accordance with the invention may use a liquid cell, may use a near-field optical element made of silicon cantilever technology, or may use an apertureless probe for apertureless near-field scanning optical microscopy. The microscope may use a tuning fork for feedback in any combination of geometries of tip, sample and tuning fork and with or without gluing of the tip to the tuning fork and with control of tip attachment or near-attachment to the tuning fork. The control of tip attachment to the tuning fork may be based on near zero backlash movement technology, and the tuning fork can be used in non-contact, contact and intermittent contact modes of operation. A fiber based feedback system may use either straight or cantilevered fibers, and the detection of signals may be based on amplitude, phase, wavelength or other optical parameters that can be used to monitor the movement of an SPM sensor.

A method for producing subwavelength near-field optical apertures includes directing a laser beam at the top of a tapered optical fiber to melt the fiber tip. The melting forms a lens at the tip, and the resulting structure is then coated with metal in such a way that an aperture is left open, or the aperture is formed in the metal by means of an ion beam or a laser beam. In another embodiment, the tip of a micropipette is melted by a laser beam to form an aperture having thick walls.

A stable, image-retaining, optically switchable film produced from a purple membrane in a high-pH polyvinyl alcohol solution forms an optical memory for data storage. The film, when dry, can be exposed to light to convert BR molecules to their M state, which is stable, and which allows long-term image storage. The image can be erased by exposing the film to yellow light to thereby switch all the molecules to the M state, and then a reverse image can be obtained using blue light. By controlling the location and wavelength of the incident light, pixels can be selected and exposed for information storage.
